No you don't have to do it in one day. Your Alcatraz tour is going to be scheduled for a specific day and time, so you have to do it on that day, but hop on hop off portion of your ticket is …

No you don't have to do it in one day. Your Alcatraz tour is going to be scheduled for a specific day and time, so you have to do it on that day, but hop on hop off portion of your ticket is more flexible. You can come back and do the bus on a different day from you Alcatraz.
